Design Notes
Sheffield 2022 designed this part for testing the function of their MutaT7-based directed evolution system ‘rEvolver’. Since this system was designed for use in V. natriegens, BBa_K4451013 has been codon-optimised accordingly. This part is used in the composite ‘MutaT7 Test Cassette’ BBa_K4451021, where chloramphenicol resistance (caused by a C → T transition mutation induced by cytosine deaminase activity) acts as a reporter of poor insulation of the hypermutation locus (see BBa_K4451021 for more information).
